  Peacock stone green is a toxic triphenylmethane chemical,which is not only a dye but also a chemical agent for killing bacteria and insects.After entering aquatic animals,peacock stone green quickly metabolizes into fat soluble colorless peacock stone green.Malachite green has potential carcinogenic,teratogenic,and mutagenic effects,and its use in the livestock industry has not been recognized by the US Food and Drug Administration(FDA);China also lists malachite green as prohibited in the agricultural industry standard NY5071-2002 Guidelines for the Use of Harmless Food Fish.Due to the lack of cost-effective alternatives,the use of malachite green in aquaculture is repeatedly prohibited.
  This product is used for rapid detection of malachite green residues in tissue samples of aquatic products(such as fish,shrimp,etc.).The entire detection process only takes about 40 minutes and is suitable for testing in various enterprises and institutions.

  Homogenizer,shaker mixer,electronic scale,weighing spoon,centrifuge,centrifuge tube,nitrogen(air)meter,pipette,timer,test tube holder,etc
  Sample processing
  1、Water sample:Use a dropper to directly aspirate the water sample for testing.
  2、Tissue like(shrimp should be thoroughly washed after turning around and shell removal,and fish should be washed after scaling)
  1.Take a certain amount of chopped tissue sample and homogenize it with a homogenizer;
  2.Weigh 3g of homogenized sample into a 15ml centrifuge tube;Add 2ml of extractant A,3ml of extractant B,and a tube of oxidant.Shake thoroughly on a vortex shaker and mix for 3 minutes,or shake by hand for 5 minutes and centrifuge at 4000r/min for 5 minutes.
  3.Take 1ml of supernatant into a 5ml centrifuge tube and add two drops of co solvent.
  4.After shaking evenly,dry with nitrogen(air)gas in a 65℃water bath.
  5.Add 0.2ml of substrate complex solution to wash the centrifuge tube(immediately proceed to step 6 after rinsing,with intervals not exceeding 3 minutes).
  6.Take 0.1ml(about three drops)of the sample solution to be tested and transfer it into the gold standard micropores.Gently blow with a special dropper for the duplicate solution until the red substance in the well is completely dissolved.Wait for 5 minutes for the reaction.
  [Usage Steps]
  1.Before testing,read the user manual completely.Before use,restore the reagent plate and sample solution to room temperature(20℃-30℃);
  2.Take out the testing card from the original packaging bag and place it horizontally in front of the observer(use immediately after opening);
  3.Take all the solutions from the gold standard micropores and add them to the sample well(see figure below).After adding the sample,start timing;
  4.The results should be read within 5-8 minutes,and interpretation at other times is invalid.
  【Result judgment】
  Negative:The color of the T line(test line,near the end of the sample well)is darker or the same as the C line(control line),indicating that the concentration of malachite green in the sample is below the detection limit or does not contain malachite green.

  Positive:The T-line appears lighter than the C-line,or the T-line appears green,indicating that the concentration to be tested in the sample is higher than the detection limit.The shallower the T-line compared to the C-line,the higher the concentration to be tested in the sample.
  Invalid:The absence of quality control line C indicates improper operation or the reagent kit has failed.In this case,carefully read the instructions again and retest with a new reagent kit.If the problem persists,immediately stop using this batch of products and contact the local supplier.
  Limitations of testing methods
  1.This product is only a qualitative detection method and its concentration cannot be determined.
  2.This product is only for preliminary screening use,and the test results cannot be used as a basis for diagnosis.Two analytical methods must be used to determine the results,and gas chromatography-mass spectrometry(GC/MS)is currently the better method for confirming analysis.

  This product has no cross reactivity with other types.
  Precision
  At the same time,this product and malachite green liquid chromatography-mass spectrometry tandem mass spectrometry were used to detect 150 samples,including 94 negative samples and 56 positive samples.The test results showed that the conformity rate between this product and mass spectrometry was 100%.
  【Precautions】
